Solvents where one or more hydrogen atoms are substituted with Deuterium atoms are termed as Deuterated Solvents. Deuterated solvents used for NMR spectroscopy.
Examples for Deuterated Solvents;
water (D2O)chloroform (CDCl3), dichloromethane (CD2Cl2), dimethyl sulfoxide (DMSO-d6), acetonitrile (CD3CN), methanol (CD3OD), acetone (acetone-d6), benzene (C6D6), toluene (toluene-d8), tetrahydrofuran (THF-d8), and trifluoroacetic acid (TFA-d).
There are three main reasons to use Deuterated Solvents in Nuclear Magnetic Resonance Spectroscopy:
A-To avoid swamping by the solvent signal.
An ordinary proton-containing solvent gives a huge solvent absorption that would dominate the 1H-NMR spectrum and interfere with the peaks of protons of interest. Most 1H- NMR spectra are therefore recorded in a deuterated solvent, because deuterium atoms absorb at a completely different frequency.
B - To stabilize the magnetic field strength.
The field strength of superconducting magnets tends to drift slowly. Modern NMR spectrometers measure the deuterium absorption of the solvent and adjust the field strength to keep the resonance frequency (field strength) constant.
C-To accurately define 0 ppm.
The difference between the deuterium frequency and 0 ppm (TMS) is well known.
To satisfy the most rigorous demands of NMR-based research and analyses, high quality, high purity, and highly enriched deuterated solvents are required. Deuterated solvent must be purified and analyzed for the presence of water content.
